Does medicine effect cycling?

I'm desperately trying to bring my nitrite down, I tried cycle which lots of people say doesnt work, I bought Stresszyme yesterday and added that, put a weird sponge thing into my filter to try and soak up the toxins, did a 10% water change and its made no difference! I've been adding melafix to the tank for the last four days, firstly because I added two guppies (to stop my lone guppy being depressed) and secondly because the tails of my guppies are starting to look torn, which happened to two of my first three guppies, eventually killing them. Is the melafix killing off my bacteria?

try doing a few major water changes over the next few days, things should settle by itself.
